GCaMP6m is stably introduced into Human Umbilical Vein Endothelial cells (HUVECs) through lentiviral transduction. Lentiviral particles that carry the GCaMP6m transgene facilitate stable genomic integration, allowing for persistent and heritable expression of the calcium indicator across multiple cell passages. To ensure uniform expression of GCaMP6m, stably transduced cells can be selected and enriched using antibiotic resistance methods (such as puromycin or hygromycin) and/or fluorescence-activated cell sorting (FACS).
