Description

The ACTOne™ MT2 stable cell line is a HEK-293 CNG derivative engineered to express recombinant human melatonin receptor 1B (MT2/MTNR1B). MT2 is primarily coupled to Gi/o proteins; receptor activation inhibits adenylyl cyclase and reduces intracellular cAMP. The assay allows both end-point and kinetic measurement of intracellular cAMP changes with a FLIPR or a fluorescence microplate reader.

About Human Melatonin Receptor 1B (MT2/MTNR1B) Stable Cell Line

Melatonin Receptor 1B (MTNR1B), commonly known as MT2, is a Class A G protein-coupled receptor (GPCR) activated by the endogenous hormone melatonin. MT2 primarily couples to Gi/o proteins, resulting in inhibition of adenylyl cyclase activity and reduced intracellular cAMP levels. The receptor is expressed in the central nervous system, retina, and several peripheral tissues, where it contributes to circadian rhythm regulation, sleep–wake timing, retinal physiology, and metabolic homeostasis.

MT2 is an important therapeutic target for insomnia, circadian rhythm disorders, mood disorders, and other conditions associated with altered melatonin signaling. MTNR1B has also attracted interest in metabolic research because genetic variation near or within the gene has been associated with fasting glucose regulation and type 2 diabetes risk.

eEnzyme's Human MT2 ACTOne™ Stable Cell Line is a HEK-293 CNG derivative engineered to express recombinant human MTNR1B. Activation of the Gi/o-coupled receptor decreases intracellular cAMP, which can be measured using the ACTOne™ fluorescent reporter system. The assay supports endpoint and kinetic measurements and is compatible with FlexStation, FLIPR, and fluorescence microplate readers.

Biological Significance of MT2

MT2 is an important mediator of melatonin signaling and contributes to the synchronization of physiological processes with environmental light–dark cycles. Through Gi/o-mediated inhibition of cAMP production, MT2 influences circadian phase regulation, sleep timing, retinal signaling, neuroendocrine function, and metabolic processes. Altered MT2 expression or activity has been investigated in sleep and circadian disorders, impaired glucose regulation, and neuropsychiatric conditions.

Because of its roles in chronobiology, nervous system function, and metabolism, MT2 remains an important target for GPCR pharmacology and drug discovery. The MT2 ACTOne™ Stable Cell Line provides a reliable platform for studying receptor function and evaluating compounds that modulate melatonin signaling.
